Cloning of Interferon in Sea Bream (Sparus aurata): An Important Marker for Immune Response after Viral Infection

چکیده
The interferons (IFNs) are soluble cytokines involved in the immune response against viral pathogens. In mammals, three families of IFNs have been identified (types I, II and III) and, recently, homologues of type I have been found in different fish species. In this paper we report the cloning of IFN genes from sea bream (Sparus aurata), expression analysis and gene structure. The sea bream IFN cDNA consists of 914 bp that translate in one reading frame to give the entire molecule containing 186 amino acids. The analysis of the sequences revealed the presence of a putative 20 amino acid signal peptide, two cysteine residues and two potential N-glycosylation sites and, finally, a high percentage of identity with type I IFN from mammals compared to type III IFN. Real time PCR was performed after poly I:C stimulation of head kidney leukocytes from sea bream to investigate the expression of sea bream IFN and Mx and an induction was observed for both genes at different time points and with different intensities. The sea bream IFN genes contains four introns, as type III IFN from mammals. The presented results will give the opportunity to investigate more in detail antiviral immune responses, vaccination and immunostimulation effects in sea bream, an economical important species for the South Mediterranean aquaculture. _____________________________________________________________________________________________________________

This work which relates to the study of growth and biometrics is an essential tool for a better monitoring of the stocks of the Gilthead sea bream Sparus aurata on the coast of Kabylia: region of Tigzirt province of Tizi-Ouzou. The results showes that the annual growth of S. aurata is 3.4 cm (all sexes combined) while the growth parameters calculated by the Von Bertalanffy equation are establis...
متن کاملExtending Immunological Profiling in the Gilthead Sea Bream, Sparus aurata, by Enriched cDNA Library Analysis, Microarray Design and Initial Studies upon the Inflammatory Response to PAMPs
This study describes the development and validation of an enriched oligonucleotide-microarray platform for Sparus aurata (SAQ) to provide a platform for transcriptomic studies in this species. A transcriptome database was constructed by assembly of gilthead sea bream sequences derived from public repositories of mRNA together with reads from a large collection of expressed sequence tags (EST) f...

The aim of the present work was to investigate the transcriptome response of gilthead sea bream (Sparus aurata) after challenge with the myxosporean Enteromyxum leei, a wide-spread enteric parasite causing heavy economic losses in Mediterranean sparid farms. This parasite causes severe desquamative enteritis which usually leads to death of the fish, and there are no preventative or curative tre...
متن کاملSuppression subtraction hybridization (SSH) and macroarray techniques reveal differential gene expression profiles in brain of sea bream infected with nodavirus.
Despite of the impact that viruses have on aquatic organisms, relatively little is known on how fish fight against these infections. In this work, the brain gene expression pattern of sea bream (Sparus aurata) in response to nodavirus infection was investigated. We used the suppression subtractive hybridization (SSH) method to generate a subtracted cDNA library enriched with gene transcripts di...

SUMMARY The cellular mucosal and systemic effectors of gilthead sea bream (GSB) (Sparus aurata) involved in the acute immune response to the intestinal parasite Enteromyxum leei were studied in fish experimentally infected by the anal route. In the intestinal inflammatory infiltrates and in lymphohaematopoietic organs (head kidney and spleen) of parasitized fish, the number of plasma cells, B c...
